The objective of this work was to analyze the agronomic performance and chromosomal stability of transgenic homozygous progenies of soybean [Glycine max (L.) Merrill.], and to confirm the resistance of these plants against Anticarsia gemmatalis. Eleven progenies expressing cry1Ac, hpt and gusA genes were evaluated for agronomic characteristics in relation to the nontransformed parent IAS 5 cultivar. Abstract The objective of this work was to analyze the androgenic response of Brazilian wheat genotypes to different pretreatments of the spikes, prior to the culture of isolated microspores, and to the effect of a gelling agent in the induction culture medium. Five genotypes were evaluated for embryo formation, green plant regeneration, and spontaneous chromosome duplication. Wheat spikes were subjected to two pretreatments: cold, at 4ºC for 21 days; and 2-hydroxynicotinic acid, at 32ºC for two days. Culture media were evaluated with or without Ficoll as a gelling agent. Association between floral bud size and developmental stage in soybean microspores BABT

This study was carried out to establish the association between floral bud size and the corresponding microspore developmental stages for Brazilian soybean cultivars. Microspore developmental stage distributions were examined in young buds from cv Década, IAS5 and RS7. The data indicated that for a given bud-size group, the microspores of different cultivars were at different developmental stages, with cv RS7 and Década distributed at the youngest and cv IAS5 at the most advanced stages. Detection of cry1 genes in Bacillus thuringiensis isolates from South of Brazil and activity against Aanticarsia gemmatalis (Lepidoptera:Noctuidae) BJM

The bacterium Bacillus thuringiensis (Bt) is characterized by its ability to produce proteic crystalline inclusions during sporulation. Cry1 protein has insecticidal activity and is highly specific to certain insects and not toxic to unrelated insects, plants or vertebrates. In this work, the patogenicity of twelve Bt isolates was tested against Anticarsia gemmatalis, one of the most important insect pests of soybeans. Evaluation of gelling agents on anther culture: response of two soybean cultivars BABT

Anthers of two soybean cultivars were cultured in B5 long basal culture media gelled with agarose or PhytagelTM. Cytological examinations of the anthers were carried out during the first 45 days of culture to assay the viability and developmental stage of microspores. Frequency of callus formation was recorded at 45 days of culture. The analysis of variance of the microspore viability assay showed significant Cultivar X Gelling Agent X Day of Culture interactions. Abstract Drought limits wheat production in the Brazilian Cerrado biome. In order to search for candidate genes associated to the response to water deficit, we analyzed the gene expression profiles, under severe drought stress, in roots and leaves of the cultivar MGS1 Aliança, a well-adapted cultivar to the Cerrado. A set of 4,422 candidate genes was found in roots and leaves. The number of down-regulated transcripts in roots was higher than the up-regulated transcripts, while the opposite occurred in leaves. The number of common transcripts between the two tissues was 1,249, while 2,124 were specific to roots and 1,049 specific to leaves. Genes de Bacillus thuringiensis: uma estratégia para conferir resistência a insetos em plantas Ciência Rural

Os insetos constituem uma das principais causas de danos à produção agrícola no mundo. O controle de insetos tem sido realizado por meio de agroquímicos e, em muito menor escala, pelo emprego de inseticidas biológicos. As plantas transgênicas resistentes a insetos representam uma nova alternativa no combate aos insetos-praga das lavouras. A bactéria entomopatogênica Bacillus thuringiensis Berlinier (Bt) é a fonte dos genes de resistência nas chamadas plantas-Bt, produzidas comercialmente. No presente trabalho de revisão, são abordados os aspectos relacionados à bactéria Bt como fonte de genes de resistência a insetos-pragas, plantas geneticamente modificadas, vantagens do uso de plantas-Bt, bem como perspectivas dessa ferramenta biotecnológica.

Induction and identification of polyploids in Cattleya intermedia Lindl. (orchidaceae) by in vitro techniques Ciência Rural

The effect of colchicine on the protocorm-like bodies of Cattleya intermedia L. (Orchidaceae) cultured in vitro was studied. Different concentrations of the drug (0.00, 0.05, 0.10 and 0.20%) as well as two times of culture (4 or 8 days) were tested to determine the best treatment for the induction of tetraploid plants. Of the 3 colchicine levels applied, 0.05 and 0.10% of drug treatments appeared to be effective on the production of mixoploids and tetraploids and could be further used in breeding programs. Influence of antibiotics on embryogenic tissue and Agrobacterium tumefaciens suppression in soybean genetic transformation Bragantia

The influence of different antibiotic treatments in soybean genetic transformation was evaluated. First, an assay was performed to verify how different antibiotic treatments affect soybean embryogenic tissues. The effect of carbenicillin at 500 mg L-1 was genotype-dependent. This antibiotic did not affect embryo survival of cv. IAS5, but a three-fold increase of embryo proliferation was observed for cv. Bragg, when compared to the control. On the other hand, cefotaxime at 350 and 500 mg L-1 caused death of embryogenic tissues of both cultivars. Finally, the association of cefotaxime (250 mg L-1) + vancomycin (250 mg L-1) did not affect negatively the somatic embryos of tested cultivars until 63 days of treatment. Initial segmentation patterns of microspores and pollen viability in soybean cultured anthers: indication of chromosome doubling BABT

Anthers obtained from flowers buds of soybean cultivar IAS-5 were cultured in two basal culture media (B5 and B5 long). Cytological examinations of the in vitro anthers were performed during the first 20 days of culture to assay the viability (by propionic-carmine and fluorescein diacetate tests) and the stage of development of pollen grains. The frequencies of viable pollen grains varied significantly between bud sizes on the propionic-carmine analysis. The basal culture media and bud size had no clear effect on the frequencies of binucleate symmetrical and multinucleate pollen grains. Chromosome counts of metaphasic microspores throughout the culture period showed microspores with higher ploidy level in addition to normal chromosome number (n=20).

Isolation and characterization of two plant growth-promoting bacteria from the rhizoplane of a legume (Lupinus albescens) in sandy soil Rev. Bras. Ciênc. Solo

Two bacterial strains that amplified part of the nifH gene, RP1p and RP2p, belonging to the genus Enterobacter and Serratia, were isolated from the rhizoplane of Lupinus albescens. These bacteria are Gram-negative, rod-shaped, motile, facultative anaerobic, and fast-growing; the colonies reach diameters of 3-4 mm within 24 h of incubation at 28 ºC. The bacteria were also able to grow at temperatures as high as 40 ºC, in the presence of high (2-3 % w/v) NaCl concentrations and pH 4 -10. Strain RP1p was able to utilize 10 of 14 C sources, while RP2p utilized nine. The isolates produced siderophores and indolic compounds, but none of them was able to solubilize phosphate. In an attempt to establish an alternative plant regeneration system for soybean [Glycine max (L.) Merrill] cultivars used in Brazilian breeding programs, ten genotypes were tested for their embryogenic potential. Cotyledons were removed as explants from immature seeds harvested from field-grown plants. After 45 days on induction medium, the number of responding cotyledons and the number of somatic embryos per immature cotyledon were evaluated. The percentage of explants that produced somatic embryos varied from 1 to 70% among cultivars. Somatic embryos of the commercial soybean (Glycine max) cultivar IAS5 were co-transformed using particle bombardment with a synthetic form of the Bacillus thuringiensis delta-endotoxin crystal protein gene cry1Ac, the beta-glucuronidase reporter gene gusA and the hygromycin resistance gene hpt. Hygromycin-resistant tissues were proliferated individually to give rise to nine sets of clones corresponding to independent transformation events. Screening of Brazilian soybean genotypes with high potential for somatic embryogenesis and plant regeneration PAB

The aim of this work was to identify Brazilian soybean (Glycine max) genotypes with potential to respond to in vitro culture stimuli for primary somatic embryo induction, secondary embryo proliferation and plant regeneration. Differences among eight tested cultivars were observed at each stage. Two cultivars, IAS-5 and BRSMG 68 Vencedora, were selected for the evaluation of the capacity for embryo differentiation and plant regeneration. Somatic embryogenesis from immature cotyledons of soybean (Glycine max (L.) Merr.): ontogeny of somatic embryos BABT

Soybean somatic embryos obtained from immature zygotic embryos were histologically analyzed in order to determine possible ontogenetic routes followed by these embryos. The proembryo stage and globular, heart-shaped, torpedo and cotyledonary embryo stages were found, closely resembling the ontogeny of zygotic embryos. However, the absence of a characteristic suspensor as well as the delay in the establishment of inner organization were the main differences between these two embryogenic processes.
